Important factors about input genomic DNA/RNA
- Human, animal, plant or microbial DNA or RNA
- Molecular weight should be more than 200 bases
- DNA/RNA can be in solution or formalin-fixed paraffin sections
- DNA/RNA can be isolated using any of the standard methods or kits
- DNA/RNA can be single-stranded or double-stranded
Important features of amplified DNA or cDNA
- Mixture of ds and ss DNA, MW ~ 500 b
- 3 – 10 micrograms DNA, ~50 micrograms/microliter
- Virtually no background (negative controls show no DNA amplification)
- Excellent representation of all sequences including GC-rich DNA
